Details

Packed with a strong anion exchanger, they can be used to fully retain acidic compounds for clean-up or to isolate them by “catch and release” process.

The acetate counter-ion is easily exchangeable (more than the chloride ion).

SiliaSep flash cartridges offer a superior alternative to the other cartridges available on the market as they present the following characteristics:

概述

该酶为 E. coli 核酸内切酶 III（Nth）的热稳定同源蛋白。既有 N-糖基化酶活性也有 AP-裂解酶活性。N-糖基化酶活性可释放双链 DNA 上受损的嘧啶碱基，产生一个脱嘧啶（AP）位点。AP-裂解酶活性随后对该位点进行切割。
Tma 核酸内切酶 III 识别 DNA 上的无碱基位点、5,6 二羟基胸腺嘧啶和胸腺嘧啶乙二醇。 

来源

重组 E. coli 菌株，基因克隆自海栖热袍菌（Thermotoga maritima）。 

反应条件

1X ThermoPol 反应缓冲液
[10 mM KCl，20 mM Tris-HCl，10 mM (NH4)2SO4，2 mM MgSO4，0.1% Trition X-100（pH 8.8 @ 25℃）]，65℃ 温育。

单位定义

1 单位指在 10 μl 反应体系中，65℃ 条件下，1 小时能切割 1 pmol 含一个 AP 位点* 的 60 mer 寡核苷酸双链所需要的酶量。单位活性检测条件请登陆 www.neb.com 或 www.neb-china.com。
*AP 位点的创建方法如下：37℃ 条件下，用 1 单位尿嘧啶-DNA 糖基化酶（UDG）处理 10 pmol 含一个尿嘧啶碱基的 60 mer 寡核苷酸双链 2 分钟。
