概述

Lambda 蛋白磷酸酶（Lambda PP）是一种 Mn2+ 依赖型蛋白磷酸酶，对磷酸化的丝氨酸、苏氨酸和酪氨酸残基均有活性。Lambda PP 也可作用于磷酸化的组氨酸残基。

来源

重组 E. coli 菌株，携带有 Lambda 噬菌体 ORF221开放阅读框（Dr.D.Barford 惠赠）。

反应条件

1X 蛋白金属磷酸酶（PMP）缓冲液 [50 mM HEPES（pH 7.5 @ 25 ℃），100 mM NaCl，2 mMDTT 和 0.01% Brij 35] 。添加 1 mM MnCl2，30℃ 温育。
热失活：在 50 mM Na2EDTA 存在的条件下，65℃ 加热 1 小时。

随酶提供的试剂

10X 蛋白金属磷酸酶（PMP）缓冲液
10X MnCl2（10 mM）

分子量

25 kDa。

质量保证

无蛋白酶污染。

单位定义

1 单位指 50 μl 反应体系中，30℃ 条件下 1 分钟内水解 1 nmol 的对硝基苯酚磷酸盐（50mM，NEB #P0757）所需要的酶量。

浓度

特性

 高效 5´ →3´ 核酸外切酶活性
 切下双链 DNA 的 5´ 单核苷酸 

概述

Lambda 核酸外切酶作用于双链 DNA，沿 5´→3´ 方向逐步切去 5´ 单核苷酸。最适底物是 5´ 磷酸化的双链 DNA，也能缓慢降解单链 DNA 和非磷酸化底物。该酶不能从 DNA 的切刻或缺口处起始消化。 

来源

E. coli 的 Lambda 核酸外切酶基因与编码麦芽糖结合蛋白（MBP）的基因融合表达。亲和层析后，从融合蛋白上切下 Lambda 核酸外切酶，并纯化除去 MBP。 

反应条件

1X Lambda 核酸外切酶反应缓冲液
[67 mM Glycine-KOH（pH 9.4 @ 25℃），2.5 mM MgCl2，50 μg/ml BSA]，37℃ 温育。
热失活：75℃ 加热 10 分钟。

注意事项

5´ -0H 端的切割速度比 5´ -PO4 端慢 20 倍。单链 DNA 比双链 DNA 慢 100 倍。
